How subverting the immune system shapes the arms race between bacteria and hosts

Why is it that Mycobacterium tuberculosis can cause tuberculosis with as little as 10 cells, whereas Vibrio cholerae requires the host to ingest up to tens of millions of cells to cause cholera? This is the question that two research teams, from the Pasteur Institute, in France, and the Instituto Gulbenkian de Ciencia and the University of Lisbon, in Portugal, answer in the latest issue of the journal PLoS Pathogens. The researchers show that bacteria that are able to invade and/or destroy cells of the host's immune system have higher infectivity, whereas those that are more motile, multiply faster and communicate with each other are less infectious, that is, it takes more bacterial cells to trigger an infection. These findings help understand the patterns that shape infectivity of bacteria, and contribute to more accurate predictions of how emerging pathogens may evolve, with implications for public health.

Despite the multitude of mechanisms that pathogens use to exploit its human host, the research teams of Eduardo Rocha, in France, and Francisco Dionsio, in Portugal, set out to find common themes in the mechanisms used by distantly related bacteria. The researchers trawled through scientific literature on 48 bacterial pathogens. They related several bacterial features (motility, interactions with the immune system, growth rate) with their value of ID50 the minimum number of bacterial cells that is required to trigger an infection in 50% of infected hosts.

The teams found that bacteria that are able to destroy or subvert phagocytes the cells of the immune system that engulf bacteria have ID50 of around 250 cells. The remaining bacteria, meanwhile, have ID50 values of around 30 million they are much less infectious. These bacteria rely on high turnover (or growth), are very motile, and are capable of communicating with other bacteria, through the mechanism of quorum-sensing.

Says Joao Gama, first author of the paper, "In the "arms race" between disease-causing bacteria and the hosts they infect, some bacteria act through frontal attack, whereas others operate through stealth, manipulating the host's immune system. Many others are somewhere in between. Our findings suggest that the low ID50 (more infectious) bacteria be classed as working by stealth; the high ID50 bacteria are those that resort to frontal attacks".

Understanding brain performance: People who take Ritalin are far more aware of their mistakes

ScienceDaily (Feb. 27, 2012) ? A new study, by Dr Rob Hester from the Department of Psychological Sciences and colleagues at the Queensland Brain Institute, investigated how the brain monitors ongoing behaviour for performance errors -- specifically failures of impulse control.

It found that a single dose of methylphenidate (Ritalin) results in significantly greater activity in the brain's error monitoring network and improved volunteers' awareness of their mistakes.

Diminished awareness of performance errors limits the extent to which humans correct their behaviour and has been linked to loss of insight in a number of clinical syndromes, including Alzheimer's Disease, Schizophrenia and Attention-Deficit Hyperactivity Disorder. The findings demonstrate that activity within those parts of the brain that deal with human error, including the dorsal anterior cingulate (dACC) and inferior parietal lobule (IPL) differs depending on whether participants are aware of their performance errors. Critically, researchers showed that a single, clinically relevant dose of methylphenidate, which works by increasing the levels of catecholamines in the brain, dramatically improved error awareness in healthy adults.

Researchers used functional magnetic resonance imaging (fMRI) to show that methylphenidate was able to promote the conscious awareness of performance errors by strengthening activation differences within the dACC and IPL for errors made with and without awareness, compared to placebo and other comparison drugs.

While the study provided only a single dose of methylphenidate to healthy participants, and needed to be replicated in people using standard clinical doses, the data highlights the potential of pharmacotherapy in addressing problems of awareness and insight that features in a range of neurologic and psychiatric conditions.

Dr Hester said failure to recognise errors was related to poor insight into a person's clinical condition, which can impair treatment.

"For example, in conditions such as Schizophrenia and Alzheimer's Disease, poor error awareness has been associated with delusions, paranoia and has been the cause of considerable distress to patients," he said.

"Failing to recognise your own error at the time can account for the difference between your recollection and the reality that confronts you. Understanding the brain mechanisms that underlie how we become conscious of our mistakes is an important first step in improving error awareness, and potentially reducing these symptoms."

Models underestimate future temperature variability: Food security at risk

ScienceDaily (Feb. 17, 2012) ? Climate warming caused by greenhouse gases is very likely to increase the variability of summertime temperatures around the world by the end of this century, a University of Washington climate scientist said Friday. The findings have major implications for food production.

Current climate models do not adequately reflect feedbacks from the relationship between the atmosphere and soil, which causes them to underestimate the increase of variability in summertime temperatures, said David Battisti, a UW professor of atmospheric sciences.

While warmer temperatures already have implications for food production in the tropics, the new findings suggest the increase in the volatility of summertime temperatures will have serious effects in grain-growing regions of Europe and North and South America, Battisti said.

"If there's greater variability, the odds of the temperature being so high that you can't grow a crop are greater," he said.

"In terms of regional and global food security, it's not good news."

Battisti presented his findings at the American Association for the Advancement of Science meeting in Vancouver, Canada. His discussion was part of a panel on climate and global food security that included Rosamond Naylor of Stanford University and Daniel Vimont of the University of Wisconsin, with whom he has collaborated on previous food security research.

Earlier research has shown that by the end of this century, the increase in average growing season temperature, if other factors remain the same, will likely reduce yields of rice, corn and soybean 30 to 40 percent. Already rice yields in the tropics are being affected by higher temperatures, affecting nations such as Indonesia, which frequently imports rice to stabilize prices, Battisti said.

Their conclusions are based on geological and other proxy records of climate and El Ni?o from the last 10,000 years, plus recent analyses of long-term climate changes because of global warming.

The Intergovernmental Panel on Climate Change, the United Nations body conducting ongoing assessments of climate change, has estimated that future month-to-month temperature variability during summer months is likely to be greater in some places and less in some places, but should stay roughly constant in many places.

But the new modeling work, Battisti said, shows most areas can expect to see greater variability in summer temperatures between now and 2085, with the biggest impacts in Europe, Africa and South America.

"The increased variability will be pretty ubiquitous. You will see it pretty much everywhere."

Increased temperature variability compounds the loss of production because of higher average temperatures, Battisti said. Add higher fertilizer prices and other market pressures to the mix "and food insecurity is likely to be higher than it has been for some time."

In Iran, a Stirring of Green Protesters Produces a Clampdown (Time.com)

There was a palpable sense of excitement among the supporters of Iran's opposition Green Movement in recent weeks. A mass rally was planned for Tuesday, the one-year anniversary of the house arrest of opposition leaders Mir Hussein Moussavi and Mehdi Karroubi, and a large turnout was expected. Flyers were posted on opposition websites and the Twitter-verse was abuzz with messages from Green supporters encouraging each other.

The Iranian regime squashed it. Even though Green supporters hit the streets in Tehran and smaller cities like Isfahan and Shiraz on Tuesday, they appeared to be outnumbered by security forces, according to accounts on opposition websites. There were also reports of arrests and sporadic clashes between riot police and demonstrators on some opposition sites. Still, the presence of demonstrators in the streets of large Iranian cities for the first time in a year shows that the Green Movement hasn't been completely snuffed out. "You can't gauge the amount of support for the opposition by the number of people in the streets. The strong crackdown by the government keeps many people away," says Hossein Bastani, an Iran analyst based in Paris. "But when there's any people at all in the streets, it still rattles the regime." In two weeks, Iran's leaders will ask voters to hit the polls for a key vote for the majlis or parliament. This will be the first election held in the country since the contested presidential election of Mahmoud Ahmadinejad in 2009, which kicked off months of violent protests. The regime was clearly concerned that Tuesday's protests could set the stage for another round of election violence and that's why they came out in force. Supreme Leader Ayatullah Ali Khamenei raised concerns about the possibility of election unrest in a Friday prayers speech two weeks ago. "The busier the ballot box and the higher the rate of participation, the greater the prestige and strength of the country," Khamenei said. And he warned of the potential pitfalls ahead. "The enemy doesn't want this. It's been two or three months that the enemy has spread propaganda to discourage people from participating in the elections. And some inside the country, without knowing what they're doing, are saying the same things. This is a careless and mercenary attitude."

Last year on Feb. 14, opposition leaders Moussavi and Karroubi called for the "25th of Bahman" protests, named after the date in the Iranian calendar, to show solidarity with the Arab Spring uprisings in the region. One of the popular slogans used by demonstrators last year was "Mubarak, Ben Ali, now it's time for Seyed Ali," a reference to Supreme Leader Khamenei. This year, the regime was well prepared. For more than a week, there has been a blackout on social networking sites like Facebook and Twitter. Even Gmail and Yahoo email were blocked. Internet speeds have almost ground to a halt. And then the intimidation campaign began: text messages and emails were sent out to students and activists threatening them with arrest if they participated in the planned demonstration on Tuesday. There was also a wave of arrests of activists in the lead-up to Tuesday's protest, according to opposition websites.

Green Movement supporters had called for demonstrators to rally in the streets of Tehran by 4 p.m. on Tuesday. Photos posted on opposition sites showed that security forces had already flooded the streets by that time. Text messaging services were also shut down. The protesters who did hit the streets in Tehran largely stayed away from controversial slogans this year, as had been advised in the demonstration flyers.

The irony is that the Iranian regime's crackdown on the "25th of Bahman" protest on Tuesday coincided with the one-year anniversary of the democratic uprising in Bahrain, a largely Shi'a-led protest movement that the Iranian government strongly supports.

"In Iran, if the security forces backed off for one day, only one day, you would see the real strength of the opposition," says Bastani. That's unlikely to happen anytime soon. In fact, the riot police are no doubt gearing up to hit the streets again for Iran's parliamentary elections on March 2.

Hollywood will meet President Obama with open wallets

Despite complaints about his failure to support Hollywood's position on online piracy, President Obama does not appear to have lost his fundraising base in the entertainment community.

As Obama arrives in Los Angeles on Wednesday, local campaign fundraisers said there has been no drop-off in Hollywood donations to his reelection bid since the D.C. demise of long-sought anti-piracy legislation. Hollywood's chief lobbyist Chris Dodd suggested last month that Obama and his fellow Democrats could pay a price for not representing the industry's interests in Washington.

But there was no evidence of that in the run-up to Wednesday's fundraising events. A dinner and a reception at the Holmby Hills home of "The Bold and the Beautiful" producer Bradley Bell and his wife, Colleen, co-hosted by actor Will Ferrell and his wife, Viveca, sold out faster than any fundraiser in the last several years, according to Ken Solomon, co-chairman of Obama's Southern California fundraising committee.

The reception, featuring an acoustic set by recent Grammy winners the Foo Fighters, will run $250 to $500 per person. A small private dinner, which will cost $35,800 a head, will follow. The two events are expected to raise more than $3 million for the campaign and Democratic National Committee.

The robust turnout underscores the ties that bind many in the entertainment industry to the Democratic president on topics such as abortion rights and the environment. Because much of Hollywood political giving is ideological, campaign donations are not usually tied to short-term legislative items, fundraisers said.

"Hollywood money for the most part is actually quite pure," said veteran Los Angeles Democratic fundraiser John Emerson, the other co-chairman of Obama's Southern California finance team. "It's given by people who really believe in the issues. They're not writing the checks because they're after some regulatory change."

Dan Glickman, who served as chairman of the Motion Picture Assn. of America from 2004 to 2010 ? the position Dodd holds now ? said some in the entertainment industry may have residual "sour feelings" from the White House's stance on the anti-piracy legislation, but not enough to make a significant dent in the president's reputation with Hollywood Democrats.

"On issues like censorship, how to deal with the less fortunate in society, dealing with the poor ? they're pretty strong advocates of the progressive agenda," Glickman said. "They're not one-issue voters."

Indeed, key figures in the entertainment community have been ardent backers of Obama's reelection bid.

Producer Paula Wagner says she has been and continues to be an Obama supporter but does not view the issue of piracy through a political lens: "This transcends politics," she said. "Nobody has the right to steal our movies, but that's not political. Maybe the bill was too Draconian and needs to be restructured without violating free speech, privacy and access to information."

Producer and former MCA Inc. President Sidney Sheinberg said Newt Gingrich once asked him why Democrats got so much Hollywood support and the Republicans did not: "I told him the reason is that most people in Hollywood vote their conscience, not their pocket book."

For example, Sheinberg said his wife, actress Lorraine Gary, "is going to vote for a pro-choice candidate."

Fundraising co-chairman Solomon, who is also chief executive of the Tennis Channel, said that even though there was some initial anger over the administration's stance on the anti-piracy measures, many top Hollywood executives now feel the initial legislation was flawed.

"A few who have been on the front line of pushing this might try to exact a pound of flesh," he said. "But the notion that steadfast supporters were going to walk out the door en masse and hand the election over to Newt Gingrich or Rick Santorum or Mitt Romney is pretty preposterous."

Some of the industry's best-known figures are raising money for the campaign. Among them are film producer Jeffrey Katzenberg and political advisor Andy Spahn, who each brought in more than $500,000 by the end of 2011, according to figures released by the campaign. Solomon also raised more than $500,000, while talent agent Ari Emanuel (brother of former White House chief of staff Rahm Emanuel) corralled between $100,000 and $200,000.

According to the Center for Responsive Politics, fundraisers working in the film, television and music industries have raised $4.75 million for Obama's election through the end of 2011 ? well above the $3.7 million raised for Obama by the same industry groups during the entire 2008 election cycle.

And when First Lady Michelle Obama visited Los Angeles two weeks ago to headline two campaign fundraisers, the sold-out events brought in more than $1 million, organizers said.

President Obama's visit is his first to Los Angeles since two bills that were strongly backed by the MPAA ? the Protect Intellectual Property Act and the more controversial Stop Online Piracy Act ? were derailed. The bills would have given the Justice Department authority to seek court orders requiring Internet search engines and payment processors to block access to websites involved in piracy.

While studios and unions argued they were necessary to combat online piracy from foreign websites, opponents including Google led a campaign to defeat the bills, viewing them as a threat to their industry and freedom of speech on the Internet.

The White House publicly criticized key aspects of the bills, angering many in Hollywood who felt the president was siding with Silicon Valley interests. Others aren't blaming the White House for the backlash against SOPA.

"Hollywood got beat because these new companies had better messaging and leveraged their extraordinary distribution and market share,'' said Charles Segars, chief executive of the arts TV channel Ovation. "I don't blame the White House for our error."

During his two-day campaign swing through California, Obama will be making stops in both Los Angeles and San Francisco, soliciting funds from Hollywood and the high-tech sector ? opposing sides in the online piracy issue.

When asked for his comment on Obama's Hollywood fundraisers, Dodd said that the MPAA hopes Obama "will use this opportunity in both communities to reiterate the important point that he made in the State of the Union: that online content theft is a problem, that it harms U.S. workers and U.S. business and that now is the time to come together to find meaningful solutions to protect American intellectual property from foreign criminals."

Hollywood donors attending Wednesday's fundraisers will likely raise the topic in person if they have the opportunity, Glickman added.

Those who "feel strongly have an obligation to bring up the issue in a cordial, courteous way," Glickman said. "And I think they probably will."
